Natural ambergris is a precious animal spice. It is a stone in the stomach of sperm whales. It is vomited or excreted by whales on the sea surface and emits a special aroma in the air for a long time. The synthesis of abromide uses perillyl alcohol as raw material and undergoes two-step oxidation with KMnO4 (ozone oxidation is used in Switzerland and sodium chromate oxidation is used in Russia). That is (1) alkaline oxidation; (2) weakly acidic oxidation; oxides are obtained. Then the oxide is soaped, dehydrated and lactonized to obtain ambroxolactone. The lactone is reduced to ambroxanediol with lithium aluminum hydride in diethyl ether (or with borane in tetrahydrofuran). Use D-camphor-β-sulfonic acid as the cyclizing agent to cyclize the diol to obtain ambrox (foreign cyclizing agents include sulfuric acid, p-toluenesulfonic acid and β-naphthalenesulfonic acid, etc.). The total yield of the whole process is 35%.
